The new HT400E is a fully automatic Solid Phase Extraction system and HPLC autosampler for unattended sample preparation and HPLC analysis.
The system is able to sequentially process up to 29 SPE tubes of 1ml or 3ml capacity. It offers multiple options for cartridge conditioning, sample loading, washing, elution, dilution, derivatization and injection.
Different syringe size options are available to handle various amounts of sample and reagents.
Based on the existing and extremely successful range of autosampler robots from Hta, the hardware is already well tested for reliability and ease of support.
A standard HT400E system is configured for four reagents plus one wash solution, one waste station and a sample injector. The modular design of the system enables multiple options for the number of reagents and wash solutions.
There are also positions for limited volume reagent vials. These are useful for adding internal standards and derivatizations.
For each sample, the tray contains one SPE cartridge position, a sample position, an elution position and one position for an extra step such as dilution or derivatization.
The control software allows easy set-up of the sample processing sequence and multiple methods can be stored and modified at any time. The flexibility of the software allows almost any methodology to be created.
Special plungers are placed in the SPE tubes that allow the syringe needle to directly and accurately deliver solution to the surface of the solid phase and eliminate any dead volume in the tube. The gives exceptional accuracy and reproducibility. The plungers also allow the unit to physically lift and move the SPE cartridge to the waste and elution stations as required.
The final sample is then injected via a built-in HPLC injector connected directly to the HPLC system. The run start/stop is controlled via contact closures, meaning the HT400E is compatible with almost all HPLC systems.
A dedicated syringe wash and waste station with its own solvent line washes both the inside and outside of the syringe.
There are many applications for automated SPE. A number of which have already been run on the system including:
• Amino Acids in CSF, Plasma & Urine
• Benzodiazepines in Serum/Plasma
• Catecholamines in Urine: Method 1 & Method 2
• Crosslink (Pyridinoline and Deoxypyridinoline) in Urine
• Metanephrines in Urine
• Tricyclic Antidepressant drugs
• t-t-Muconid Acid in Urine
• VMA, 5-HIAA, HVA in Urine: Method 1 & Method 2
